See the immune repertoire in context with ImmunoSightTM
Expand the functional relevance of immune repertoire sequencing data with a combined quantitative gene expression immunophenotyping panel
To fully understand the immune response to disease and advance therapeutic development, it is crucial to sequence the Adaptive Immune Receptor Repertoire (AIRR) and quantify adaptive immune cell gene expression. However, using whole transcriptome RNA-seq to characterize the expression of key immune gene markers results in complicated bioinformatics analysis and decreased sensitivity.
At iRepertoire, we have developed ImmunoSight, a fully customizable, targeted NGS assay that quantifies the expression of 150+ immunophenotype genes alongside AIRR sequence data, at bulk or single-cell resolution, to provide biopharmaceutical researchers with the comprehensive data needed to move forward their clinical development and drug discovery programs.
Advancing key clinical areas with multi-chain repertoire analysis
Optimize cell therapy manufacturing and QC
Analyze immunophenotype-specific genes as part of your manufacturing release criteria.
Characterize the Tumor Micro-environment (TME) in-depth
Extract reliable TME gene expression data from clinical samples, including FFPE-derived RNA, to assess the impact of TME markers on immunotherapy efficacy.
Discover biomarkers of therapeutic importance
Measure patient immune response during clinical trials and reveal biomarkers indicative of therapeutic safety and efficacy.
Uncover adaptive immune cell phenotypes with customizable targeted gene amplification
ImmunoSight leverages oligo-dT reverse RNA transcription using UMI-enabled dam-PCR chemistry and gene-specific primers for 150+ genes carefully selected to characterize a wide range of immune phenotypes along with the AIRR.
ImmunoSight features:
- Adaptable: Compatible with single-cell and bulk-cell (PBMC, PAXgene RNA, sorted cells) and tissue (FFPE, FF).
- Relevant: Essential and customizable immunophenotype content (150+ genes).
- Intuitive: Quantitative relative gene expression data presented in easy-to-interpret heat maps.
Receive targeted immunophenotyping data with AIRR insights
iRepertoire’s ImmunoSight companion assay to our AIRR sequencing service provides adaptive immune receptor chain diversity and clonality data (I), coupled with targeted gene expression information (II).
In the featured data example, Panel I show tree maps denoting the relative frequency of unique V-J-uCDR3s obtained using our AIRR sequencing service. After isolating the starting material (A), cells were processed to promote the expansion of specific T cell clones (B). As shown in Panel I, the process yields the expansion of specific T cell clones (see turquoise clone expansion). Using Immunosight on the same reaction, immunophenotyping was performed, and it was found that the expanded cell product had increased relative expression of important cellular activation markers across multiple donors (Panel II).
